    Analyzing Conflict Freedom For Multi-threaded Programs With Time Annotations

    Avoiding access conflicts is a major challenge in the design of multi-threaded programs. In the context of real-time systems, the absence of conflicts can be guaranteed by ensuring that no two potentially conflicting accesses are ever scheduled concurrently.In this paper, we analyze programs that carry time annotations specifying the time for executing each statement. We propose a technique for verifying that a multi-threaded program with time annotations is free of access conflicts. In particular, we generate constraints that reflect the possible schedules for executing the program and the required properties. We then invoke an SMT solver in order to verify that no execution gives rise to concurrent conflicting accesses.     Ensuring Average Recovery with Adversarial Scheduler

    In this paper, we focus on revising a given program so that the average recovery time in the presence of an adversarial scheduler is bounded by a given threshold lambda. Specifically, we consider the scenario where the fault (or other unexpected action) perturbs the program to a state that is outside its set of legitimate states. Starting from this state, the program executes its actions/transitions to recover to legitimate states. However, the adversarial scheduler can force the program to reach one illegitimate state that requires a longer recovery time. To ensure that the average recovery time is less than lambda, we need to remove certain transitions/behaviors. We show that achieving this average response time while removing minimum transitions is NP-hard. In other words, there is a tradeoff between the time taken to synthesize the program and the transitions preserved to reduce the average convergence time. We present six different heuristics and evaluate this tradeoff with case studies. Finally, we note that the average convergence time considered here requires formalization of hyperproperties. Hence, this work also demonstrates feasibility of adding (certain) hyperproperties to an existing program

    Red Flags, Red Herrings, and Common Ground: An Expert Study in Response to State Reading Policy

    In many U.S. states, legislation seeks to define effective instruction for beginning readers, creating an urgent need to turn to scholars who are knowledgeable about ongoing reading research. This mixed-methods study considers the extent to which recognized literacy experts agreed with recommendations about instruction that were included on a state’s reading initiative website. Our purpose was to guide implementation and inform policy-makers. In alignment with the initiative, experts agreed reading aloud, comprehension, vocabulary, fluency, phonological awareness, and phonics all deserve a place in early literacy instruction. Additionally, they agreed some components not included on the website warranted attention, such as motivation, oral language, reading volume, writing, and needs-based instruction. Further, experts cautioned against extremes in describing aspects of early reading instruction. Findings suggest that experts’ knowledge of the vast body of ongoing research about reading can be a helpful guide to policy formation and implementation

    The Optimized Transport Scheme of Empty and Heavy Containers with Novel Genetic Algorithm

    To design the transport scheme of empty and heavy containers reasonably, a model with objective maximizing the route benefits is proposed. The model considered two factors: (1) the fluctuation of cargo transport demand and the switching of different voyages; (2) the optimal transport scheme of empty and heavy containers in slack and brisk seasons and the handover process of these two seasons. In order to solve this model, a novel GA is developed. With this model and algorithm, the optimal transport scheme of empty and heavy containers is put forward, and the optimization allocation of resources can be realized. The case study about China-Europe route proves that this model can improve the liner company's benefits effectively

    Ku80 cooperates with CBP to promote COX-2 expression and tumor growth.

    Cyclooxygenase-2 (COX-2) plays an important role in lung cancer development and progression. Using streptavidin-agarose pulldown and proteomics assay, we identified and validated Ku80, a dimer of Ku participating in the repair of broken DNA double strands, as a new binding protein of the COX-2 gene promoter. Overexpression of Ku80 up-regulated COX-2 promoter activation and COX-2 expression in lung cancer cells. Silencing of Ku80 by siRNA down-regulated COX-2 expression and inhibited tumor cell growth in vitro and in a xenograft mouse model. Ku80 knockdown suppressed phosphorylation of ERK, resulting in an inactivation of the MAPK pathway. Moreover, CBP, a transcription co-activator, interacted with and acetylated Ku80 to co-regulate the activation of COX-2 promoter. Overexpression of CBP increased Ku80 acetylation, thereby promoting COX-2 expression and cell growth. Suppression of CBP by a CBP-specific inhibitor or siRNA inhibited COX-2 expression as well as tumor cell growth. Tissue microarray immunohistochemical analysis of lung adenocarcinomas revealed a strong positive correlation between levels of Ku80 and COX-2 and clinicopathologic variables. Overexpression of Ku80 was associated with poor prognosis in patients with lung cancers. We conclude that Ku80 promotes COX-2 expression and tumor growth and is a potential therapeutic target in lung cancer

    Endothelial cell-specific deletion of a microRNA accelerates atherosclerosis

    Background and aims: Chronic vascular endothelial inflammation predisposes to atherosclerosis; however, the cell-autonomous roles for endothelial-expressing microRNAs (miRNAs) are poorly understood in this process. MiR-181b is expressed in several cellular constituents relevant to lesion formation. The aim of this study is to examine the role of genetic deficiency of the miR-181b locus in endothelial cells during atherogenesis. Methods and Results: Using a proprotein convertase subtilisin/kexin type 9 (PCSK9)-induced atherosclerosis mouse model, we demonstrated that endothelial cell (EC)-specific deletion of miR-181a2b2 significantly promoted atherosclerotic lesion formation, cell adhesion molecule expression, and the influx of lesional macrophages in the vessel wall. Yet, endothelium deletion of miR-181a2b2 did not affect body weight, lipid metabolism, anti-inflammatory Ly6Clow or the pro-inflammatory Ly6Cinterm and Ly6Chigh fractions in circulating peripheral blood mononuclear cells (PBMCs), and pro-inflammatory or anti-inflammatory mediators in both bone marrow (BM) and PBMCs. Mechanistically, bulk RNA-seq and gene set enrichment analysis of ECs enriched from the aortic arch intima, as well as single cell RNA-seq from atherosclerotic lesions, revealed that endothelial miR-181a2b2 serves as a critical regulatory hub in controlling endothelial inflammation, cell adhesion, cell cycle, and immune response during atherosclerosis. Conclusions: Our study establishes that deficiency of a miRNA specifically in the vascular endothelium is sufficient to profoundly impact atherogenesis. Endothelial miR-181a2b2 deficiency regulates multiple key pathways related to endothelial inflammation, cell adhesion, cell cycle, and immune response involved in the development of atherosclerosis

    Prevalence and Prognostic Significance of HPV in Laryngeal Squamous Cell Carcinoma in Northeast China

    Background/Aims: Human papillomavirus (HPV) is an etiological risk factor for a subset of head and neck squamous cell carcinomas. HPV has been proven to be a powerful prognostic biomarker for oropharyngeal cancer, but its role in the larynx has not been explored in depth. Here, we sought to evaluate the prevalence and genotype distribution of HPV in patients with laryngeal squamous cell carcinoma (LSCC) in northeast China. Methods: HPV DNA in specimens from 211 patients diagnosed with LSCC was analyzed by the polymerase chain reaction and in situ hybridization, and p16 overexpression was evaluated by immunohistochemistry. p16 expression was scored positive if strong and diffuse nuclear and cytoplasmic staining was present in > 75% of tumor cells. Results: In this study, infection with HPV and p16 expression were not absolutely consistent. Among all patients, 132 (62.6%) were positive for HPV DNA (HPV+), while 23 (10.9%) were inconsistent for HPV and p16. Multivariate analysis indicated that HPV, but not p16, is an independent prognostic factor for overall survival in LSCC. Overall survival was significantly improved in HPV+ LSCC patients compared with the HPV-negative group (hazard ratio, 0.395; 95% confidence interval, 0.185–0.843; p = 0.016). Among the 132 HPV+ patients, 28 (21.2%) were HPV-16 single infection. Conclusion: This study indicates that HPV DNA is a more reliable surrogate marker than p16 for the prediction of survival in patients with LSCC
